I had to uninstall Endpoint Upgrade Assistant in order to install epo 5.10 update 10. Will this cause any problem? or it doesnt matter?

Afaik, This is a kind of Extension which gets checked-in your ePO for upgrading the environment typically from VSE to ENS.

So I don't think you will see any major issue.

But as recommended please have the ePO and ePO Databse backed up , If in case something goes wrong or you misses something in terms of Endpoint Upgrade Assistant then you can restore it,

It is not needed and won't impact your environment.  As advised, make sure you have good backups.
